me and my team facing seperation issue on determining the N-Nitroso N-desethyl sunitinib in Sunitinib capsules by lcmsms and we are tried all kind of column chemistry and mobile phase and pH combination even though we are unable to seperate the API and Standard Analyte can anyone suggest me further proceedings
What have you tested so far? Have you achieved at least a partial separation?
If you can achieve a partial separation on two columns with different chemistries, you could try coupling them and see how it performs.
Also, the API and the nitroso impurity do not necessarily need to be baseline separated. If the sample concentration is not too high (or if your acceptable intake limit is relatively high), ion suppression and recovery might already be within an acceptable range.
Of course, this depends on the data, and I do not have enough information to draw a firm conclusion.
Maybe you could share some chromatograms and details of the experiments you’ve already performed. That would make it much easier for the community to provide useful suggestions.
