Nitrosamine toxicity testing using Caco-2 cell lines

The fact that N-nitroso compounds (NOC) are genotoxic, carcinogenic to animals, and may play a role in human cancer development.
It was also described that nitrosamides had little effect on gene expression when tested on Caco- 2 cell lines whereas nitrosamines had a strong imact on the transcriptomic profile.
Four nitrosamines, N-nitrosodiethylamine (NDEA, 50mM), N-nitrosodimethylamine (NDMA, 100mM), N-nitrosopiperidine (NPIP, 40mM), and N-nitrosopyrrolidine (NPYR, 100mM), were studied in the human colon carcinoma cell line Caco-2.
Whilst the above concentrations were tested in vitro and considered as high incubation concentrations chronic exposure to lower concentrations delivered by , for example 1000 mg metformin ( two or three tablets a day) in diabetic patients can probably reach genotoxic levels on lifetime exposal to these nitrosamines
